The callus obtained from cultured explants is sliced and teased apart into small pieces which are then incubated in a liquid medium. Once a good embryogenic suspension has been established, somatic embryos can be obtained either by allowing the culture to age or by incubating the embryogenic tissue in a medium without 2,4-D. Since an unspplemented basal medium often encourages root formation, the normal practise is to add 500ml glutamine with 0.1 µm ABA to the medium to facilitate the embryo development.
